Drug-induced mitochondrial injury can be caused by many different mechanisms including inhibition of mitochondrial DNA replication, transcription, translation, and altered protein function. Determination of the level of mitochondrial DNA relative to the nuclear DNA levels provides important information on potential mitochondrial toxicity.

Nutrient status shapes selfish mitochondrial genome dynamics across different levels of selection

It seems that you're in Germany. We have a dedicated site for Germany. This third edition is comprised of well-established protocols that are considered the gold standard in the field as well as new methodologies for mitochondrial DNA analysis. Written for the Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols and tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Mitochondrial DNA: Methods and Protocols, Third Edition will be useful not only for mitochondrial researchers but also for scientists studying human diseases where mitochondrial DNA variation has been recognized as an important pathogenic factor, including cancer and neurodegeneration.

The Human Mitochondrial Genome: From Basic Biology to Disease offers a comprehensive, up-to-date examination of human mitochondrial genomics, connecting basic research to translational medicine across a range of disease types. Here, international experts discuss the essential biology of human mitochondrial DNA mtDNA , including its maintenance, repair, segregation, and heredity. Furthermore, mtDNA evolution and exploitation, mutations, methods, and models for functional studies of mtDNA are dealt with. Disease discussion is accompanied by approaches for treatment strategies, with disease areas discussed including cancer, neurodegenerative, age-related, mtDNA depletion, deletion, and point mutation diseases. With increasing funding for mtDNA studies, many clinicians and clinician scientists are turning their attention to mtDNA disease association. This book provides the tools and background knowledge required to perform new, impactful research in this exciting space, from distinguishing a haplogroup-defining variant or disease-related mutation to exploring emerging therapeutic pathways. Active researchers in biochemistry, molecular biology, cell biology, oncology, neurology, neuroscience, and pharmacology; clinician scientists.

This form of genetic analysis can be applied to human, animal, and plant specimens. Ancient DNA can be extracted from various fossilized specimens including bones, eggshells, and artificially preserved tissues in human and animal specimens. In plants, Ancient DNA can be extracted from seeds and tissue. Archaeogenetics provides us with genetic evidence of ancient population group migrations, [1] domestication events, and plant and animal evolution. Archaeogenetics receives its name from the Greek word arkhaios , meaning "ancient", and the term genetics , meaning "the study of heredity". In February , scientists reported, for the first time, the sequencing of DNA from animal remains , a mammoth in this instance, over a million years old, the oldest DNA sequenced to date.

Likewise, our understanding of the basic biology of somatic mtDNA mutations continues to improve. In Mitochondrial DNA: Methods and Protocols, Second Edition.

The Human Mitochondrial Genome

Cooperation and cheating are widespread evolutionary strategies. While cheating confers an advantage to individual entities within a group, competition between groups favors cooperation. Selfish or cheater mitochondrial DNA mtDNA proliferates within hosts while being selected against at the level of host fitness. How does environment shape cheater dynamics across different selection levels? Focusing on food availability, we address this question using heteroplasmic Caenorhabditis elegans. We find that the proliferation of selfish mtDNA within hosts depends on nutrient status stimulating mtDNA biogenesis in the developing germline.

In most mammalian cells, mitochondria generate the bulk of ATP required to sustain a plethora of diverse cellular processes. Besides generating ATP, mitochondria also play important roles in intracellular calcium signalling [ 1 ], apoptosis [ 2 ], reactive oxygen species ROS production [ 3 ], biosynthesis of heme and iron-sulphur clusters [ 4 , 5 ], and other cellular processes.

Mitochondrial DNA copy number mtDNA-CN , a measure of the number of mitochondrial genomes per cell, is a minimally invasive proxy measure for mitochondrial function and has been associated with several aging-related diseases. Mitochondrial dysfunction has long been known to play an important role in the underlying etiology of several aging-related diseases, including cardiovascular disease CVD , neurodegenerative disorders and cancer[ 1 ].
